  • regular physical exercise – at least 150 minutes of moderate-intensity activity or at least 75 minutes of vigorous-intensity activity per week
  • Active social contact (participation in meetings or attending parties, visiting friends or relatives, traveling, and chatting online) – at least twice per week
  • active cognitive activity (writing, reading, playing cards, mahjong, and other games) – at least twice per week
  • never smokes (participants who had smoked less than 100 cigarettes in their lifetime) or used to smoke (participants who had quit smoking at least 3 years before the study)
  • never drank alcohol or drank occasionally

Researchers categorized participants into groups depending on their number of healthy lifestyle factors:

  • 0-1 healthy lifestyle factors = unfavorable lifestyle (6967 participants)
  • 2-3 healthy lifestyle factors = average lifestyle (16,549 participants)
  • 4-6 healthy lifestyle factors = favorable lifestyle (5556 participants)

All participants’ mean memory test scores declined continuously over the decade, consistent with how memory declines with age. However, the highest memory test scores were observed in the favorable lifestyle group and the lowest in the unfavorable lifestyle group, indicating that participants with favorable lifestyles had slower memory decline than those with unfavorable lifestyles (by 0.028 points/year).

The results showed that a healthy diet had the strongest effect on memory, followed by an active cognitive activity, regular physical exercise, active social contact, never or former smoking, and never drinking.

They were asked to provide information about their reproductive history, including the age of puberty onset, the age at which menopause started, and the number of pregnancies they had. They also provided information on their use of contraceptive pills.

The women were then split into four groups according to their reproductive lifespan (RL), or the number of years between puberty onset and menopause. Those in the shortest RL group had up to 31 reproductive years, whereas those in the longest RL group had 36 years or more.

The researchers also collected data on stroke incidence. During the study, they recorded 15,139 cases of new-onset stroke.

The researchers found that 13.2{cfdf3f5372635aeb15fd3e2aecc7cb5d7150695e02bd72e0a44f1581164ad809} of women in the longest RL group had a stroke, compared to 12.6{cfdf3f5372635aeb15fd3e2aecc7cb5d7150695e02bd72e0a44f1581164ad809} of women in the shortest RL group.

However, after adjusting for factors including smoking, age, and physical activity, they found that, in fact, participants in the longest RL group had a 5{cfdf3f5372635aeb15fd3e2aecc7cb5d7150695e02bd72e0a44f1581164ad809} lower risk for all types of stroke than those in the shortest group.

In particular, when they looked at different stroke subtypes, they found that those in the longest RL group had a 13{cfdf3f5372635aeb15fd3e2aecc7cb5d7150695e02bd72e0a44f1581164ad809} lower risk for intracerebral hemorrhage compared to those in the shortest RL group. Intracerebral hemorrhage is the most fatal kind of stroke which occurs when blood vessels burst in the brain.

“Preclinical studies have shown estrogen to be a profound neuroprotective agent, while clinical trials of estrogen replacement therapy in postmenopausal women failed to show benefit,” she noted.

Dr. Alex Polyakov, associate professor and gynecologist at the University of Melbourne in Australia, also not involved in the study, agreed that the exact mechanisms behind estrogen’s effects on stroke risk are not fully understood.

He added that: “Further research is needed to better understand the biological, behavioral, and social factors that may contribute to the link. Some potential mechanisms include the ability of estrogen to influence blood flow and the formation of blood clots, which can play a role in stroke risk.”

The hearts obtained from ischemic heart failure patients showed lower expression levels of BPIFB4 in the cardiomyocytes and endothelial cells than individuals with healthy hearts. The hearts of ischemic failure patients also had lower capillary density than healthy hearts.

Moreover, there was a reduction in the density of pericytes and lower coverage of blood vessels by pericytes in hearts obtained from ischemic heart failure patients.

Among ischemic heart failure patients, individuals who expressed the LAV variant of BPIFB4 on both chromosomes showed a higher density of pericytes and greater coverage of capillaries by pericytes than those who carried a single or no copies of the LAV-BPIFB4 gene.

However, these groups did not differ in capillary density, suggesting that LAV-BPIFB4 preserves the coverage of capillaries by pericytes without influencing the capillary density.

The pericytes isolated from the hearts of ischemic heart failure patients showed greater expression of markers of oxidative stress and cellular senescence. Senescence refers to the permanent arrest of cell growth and is observed in response to cellular stress and aging.

The researchers found that the addition of LAV-BPIFB4, but not other forms of BPIFB4, to cultured pericytes isolated from the heart of ischemic heart failure patients reduced the expression of senescence and oxidative stress markers.

Pericytes from the hearts of ischemic failure patients also showed a downregulation of the protein synthesis machinery, which is associated with cellular aging. The addition of LAV-BPIFB4 led to the upregulation of the protein synthesis machinery in the pericytes isolated from the hearts of ischemic heart failure patients.

Previous studies have shown that endothelial cell senescence is associated with impaired angiogenesis. In subsequent experiments conducted using endothelial cell cultures, LAV-BPIFB4 enhanced the ability of senescent endothelial cells to form new blood vessels to a greater extent than other BPIFB4 variants.

LAV-BPIFB4 also increased the secretion of molecules that promote angiogenesis by pericytes isolated from the hearts of patients with ischemic heart failure.

Preincubation of pericytes from ischemic heart failure patients with LAV-BPIFB4 also enhanced the ability of these pericytes to support the formation of new blood vessels by senescent endothelial cells.

These results suggest that LAV-BPIFB4 could directly enhance the ability of senescent endothelial cells to form new blood vessels as well as indirectly by promoting the secretion of molecules by pericytes that act on endothelial cells. In other words, the LAV-BPIFB4 gene could help restore deficits in the function of aged vascular cells.
